Re-polarisation of Macrophages Within Collective Tumour Cell Migration: A Multiscale Moving Boundary Approach
نویسندگان
چکیده
Cancer invasion of the surrounding tissue is a multiscale process collective cell movement that involves not only tumour cells but also other immune in environment, such as tumour-associated macrophages (TAMs). The heterogeneity these cells, with two extremes being pro-inflammatory and anti-tumour M1 anti-inflammatory pro-tumour M2 has significant impact on cancer interact different ways ExtraCellular Matrix (ECM). Experimental studies have shown co-migrate TAMs, TAM sub-populations (which can change their phenotype re-polarise depending microenvironment) this co-migration fully understood. In study, we extend previous multi-scale moving boundary mathematical model, by introducing M1-like alongside exerted effects process. With help model investigate numerically re-polarising TAMs into anti-tumoral how strategy affects overall progression. particular, whether M2?M1 re-polarisation could depend time and/or space, what would be macroscopic spatial- temporal-dependent invasion.

Multiscale Cues Drive Collective Cell Migration
To investigate complex biophysical relationships driving directed cell migration, we developed a biomimetic platform that allows perturbation of microscale geometric constraints with concomitant nanoscale contact guidance architectures. This permits us to elucidate the influence, and parse out the relative contribution, of multiscale features, and define how these physical inputs are jointly pr...

Tumour-associated macrophage polarisation and re-education with immunotherapy.
Monocytes/macrophages constitute important contributors of cancer-associated inflammation. Through their plasticity and capacity to become polarised by tumours towards less activatory and more immunosuppressive (M2) phenotypes, tumour-associated macrophages (TAM) are thought to support tumour progression. Orchestrated by T helper 2 (Th2)-biased stimuli, macrophage recruitment, activation and po...
متن کاملCollective guidance of collective cell migration.
Some cells migrate and find their way as solitary entities. However, during development of multicellular animals and possibly during tumor dissemination, cells often move as groups, associated tightly or loosely. Recent advances in live imaging have aided examination of such 'multicellular cell biology'. Here, I propose a model for how a group of cells can process and react to guidance informat...
متن کاملCollective cell migration in development
During embryonic development, tissues undergo major rearrangements that lead to germ layer positioning, patterning, and organ morphogenesis. Often these morphogenetic movements are accomplished by the coordinated and cooperative migration of the constituent cells, referred to as collective cell migration. The molecular and biomechanical mechanisms underlying collective migration of developing t...
